I am not sure, but does it have POS on V8's?

The standard 4.2 V8 on S6 has ignition distributors. Not sure about the S6+, maybe it has too.

You're having misfires on one bank, first check if it has a distributor in the back of the misfiring bank, if yes, then check the rotor and the cap. If those are fine, you go higher up the chain - spark wires, plugs, ignition coils.

      *EDIT*
Nope, the S6+ has coil-on-plug and YES, it has POS's, one for each bank. Go for the POS first.

The S6plus is indeed coilpack on plug but unlike the later V8s, it has external POSs, not integral internal POSs. In fact the POSs are the same as the early A4 1.8T POSs, i.e. the input side has 5 pins and the out put side has 4 pins. The AHK just has two of these. http://audi.7zap.com/en/rdw/audi+s6+.../905-905035/#1

PN is 4D0905351 Huco and Bosch both make them. https://www.ecstuning.com/Audi-B5_A4...ews/ES2515501/

urS4boy : Thank you very much for so informative answer .

I have changed Huco replacement today but without bigger effect . Car run smoothly but maybe tomorow it will be the same as yesterday .
I had problem with injectors , because autogas system was in the car before and i think another one injector is gone.
Do you have original number of injectors ?

it is likely that the injector harness was cut and spliced to LPG ecu. Typically those jobs are not done properly.

I know what you mean , but this harness was made with conectors without cutting original wires . This was first thing I checked

do a compression check. I bet it's a tooth out on cam timing. I had an RS4 do the same.
