Ok, now you're talking, I'm pretty good with a multimeter! Any ideas what sort of voltage I should get? I'm guessing 12v?

Not sure but just test one that is known to be working and use that as a marker

You could try an LED in the injector connector. It should flash with the injector pulse if you have a connection.

            So...

...I'm back, and I'm still running on 4 cylinders

In addition to all the previous checks, I've now tried:

            - Checking the voltage on injector No. 2. Its producing 14v just like the others. No problem there.

            - Testing the cylinder pressures: 1=11.8 Bar, 2=12.1 Bar, 3=11.9 Bar, 4=11.2 Bar and 5=11.9 Bar. If anything the cylinder in question had a higher pressure than the others.

            Any other suggestions, please

When did the fault first start was it anything that trigger it off?

Did you check for a spark on cylinder 2?

I parked the car up outside some shops for around 15 mins. Got back in started it up and found it running lumpy. It was running smoothly before. No warnings at all.

Did you check for a spark on cylinder 2?
No, not yet. Do I just plug the spark plug into the coil pack and check for a spark when turning the engine over?

have you done everything stated here?
                      http://www.sjmautotechnik.com/troubl...4_Ignition.htm
Yeah, I've read through it a few times and tried everything except replacing the fuel filter and checking if I'm getting a spark. Didn't think it would be the fuel filter because i've only got a problem on cylinder 2. Maybe I should change it anyway.

have you ran vag.com on the car to see if it has shown any codes?
I don't have vag.com, is that something I can buy from VAG parts?

Yes. Just make sure you rest the threaded part of the plug to a ground point, like the head.
