Swap chipset from ecu that is now blowing the fuse (now bust?) into the other ecu
check timing is 100% (front end off to turn engine over) (or any other suggestions for turning the engine?)
Check crank mark
check cam pulley
check cam marks
check dizzy alignment.
Andy,
Dont think eproms would take out S27 20a fuse.
As twoqu suggested have you tried unplugging all injectors and turning on Ignition?
To check the ECU I would remove as many sensors as possible fairly easy job with plugs
Injectors, Ignition coil, maf etc
Then see if you can connect vagcom to ECU without fuse S27 fail.
If so start to reconnect sensors one at a time and maybe find what is killing fuse S27
Timing would prevent engine start but it does not blow fuses.

Without doubt the car will not start without 12V to the injectors via the fuel pump relay and ecu with ign on.

Tried unplugging
injectors
maf
tps
coil
4 plugs on firewall (crank sensor etc)
2 aux looms
Did you swap back or try removing fuel pump relay as this could blow fuse S27 if your "new" relay coil was faulty?
I ask as prior to changing the relay you only had a problem with S28?
